Description:

Vonaspire 20 mg is a prescription oral medicine containing vonoprazan, a first-in-class potassium-competitive acid blocker (P-CAB) used for the treatment and prevention of acid-related gastrointestinal diseases. It provides fast, strong and sustained inhibition of gastric acid secretion by selectively blocking the H+ K+-ATPase (proton pump) in gastric parietal cells. Vonaspire 20 mg film-coated tablets are indicated for gastric and duodenal ulcers, reflux esophagitis, and for the prevention of ulcer recurrence in patients receiving low-dose aspirin or non-steroidal anti-inflammatory drugs (NSAIDs). It is also used as part of combination therapy for Helicobacter pylori eradication in specific clinical situations. This product is intended for adult patients under medical supervision.


Indications:

  • Treatment of gastric ulcer.
  • Treatment of duodenal ulcer.
  • Treatment of reflux esophagitis (gastroesophageal reflux disease with esophageal inflammation).
  • Prevention of recurrence of gastric or duodenal ulcer during low-dose aspirin therapy.
  • Prevention of recurrence of gastric or duodenal ulcer during non-steroidal anti-inflammatory drug (NSAID) therapy.
  • Adjunct to Helicobacter pylori eradication therapy in the following settings: gastric ulcer, duodenal ulcer, gastric mucosa-associated lymphoid tissue (MALT) lymphoma, idiopathic thrombocytopenic purpura, post-endoscopic resection of early-stage gastric cancer, or Helicobacter pylori gastritis.
  • Relief of symptoms associated with excess stomach acid such as heartburn, acid regurgitation, and epigastric pain, when related to the above conditions.

Active Ingredients / Key Components:

  • Vonoprazan 20 mg (as vonoprazan fumarate) per film-coated tablet.

Dosage & Administration / How to Use:

  • Use exactly as prescribed by your physician. Do not exceed the recommended dose or duration.
  • Typical adult dose for gastric or duodenal ulcer and reflux esophagitis is often 20 mg once daily; however, exact dosing regimen, treatment duration, and any dose adjustments must be determined by the treating physician based on the patient’s condition and response.
  • For prevention of ulcer recurrence during low-dose aspirin or NSAID therapy, the dose and duration are individualized by the physician.
  • For Helicobacter pylori eradication, Vonaspire 20 mg is used as part of a combination regimen with appropriate antibiotics; follow the exact triple therapy or combination schedule prescribed by your doctor.
  • Swallow the film-coated tablet whole with a sufficient amount of water. Do not crush, chew, or split the tablet.
  • Vonaspire 20 mg may be taken with or without food, according to your doctor’s instructions.
  • If you miss a dose, take it as soon as you remember unless it is almost time for the next dose. Do not double the dose to make up for a missed dose.
  • Do not stop treatment abruptly without consulting your doctor, even if symptoms improve, as this may lead to relapse of the condition.

Warnings & Contraindications:

  • Contraindicated in patients with known hypersensitivity to vonoprazan or any component of the formulation.
  • Do not use without medical supervision if you have severe liver impairment, severe kidney disease, or a history of gastric malignancy, unless your doctor has evaluated you.
  • Long-term suppression of gastric acid may be associated with increased risk of certain infections (such as gastrointestinal infections) and possible nutrient malabsorption; your doctor will monitor you if long-term therapy is needed.
  • Use with caution in patients with osteoporosis or risk factors for fractures, or those on long-term acid suppression, as directed by a physician.
  • Inform your doctor about all medicines you are taking, including anticoagulants, antiplatelets, antifungals, antivirals, and other acid-suppressing agents, to avoid potential drug interactions.
  • Consult your doctor before use if pregnant or planning pregnancy. The safety of vonoprazan in pregnancy and breastfeeding is not fully established; use only if the potential benefit justifies the potential risk.
  • Consult your doctor before use if breastfeeding; a decision should be made whether to discontinue nursing or discontinue the drug, considering the benefit to the mother.
  • Consult your doctor before use if you have liver disease, kidney disease, osteoporosis, electrolyte disturbances, or any serious chronic illness.
  • Seek medical advice if you experience alarm symptoms such as unintentional weight loss, persistent vomiting, difficulty swallowing, vomiting blood, or black/tarry stools.
  • Not intended to diagnose, treat, cure, or prevent any disease without proper medical evaluation and prescription.
  • Keep out of reach of children.

Side Effects:
  • Common side effects may include headache, abdominal pain, diarrhea, constipation, nausea, vomiting, flatulence, or mild indigestion.
  • Some patients may experience dizziness, fatigue, or skin reactions such as rash or itching.
  • Less common but potentially serious side effects can include severe allergic reactions (swelling of the face, lips, tongue, or throat; difficulty breathing), severe abdominal pain, persistent diarrhea, or signs of liver problems (yellowing of the skin or eyes, dark urine). Seek immediate medical attention if these occur.
  • Long-term use of potent acid-suppressing therapy may be associated with changes in certain blood tests (such as magnesium, vitamin B12, or other electrolytes); your doctor may monitor these if prolonged therapy is required.
